| Licensed for PCR* Pwo DNA Polymerase is a highly processive 5 ½ 3 DNA polymerase with additional 3 ½ 5 exonuclease (proof-reading) activity. Pwo DNA Polymerase was originally isolated from the thermophilic archaebacterium Pyrococcus woesei; GENAXIS Biotechnology Pwo DNA Polymerase is the recombinant form cloned in E. coli. The enzyme has no detectable 5 ½ 3 exonuclease activity. Pwo DNA Polymerase exhibits increased thermal stability as well as a tenfold increase in fidelity of DNA synthesis compared to Taq DNA Polymerase. Pwo DNA Polymerase generates blunt-ended PCR* products. APPLICATIONS:

UNIT DEFINITION:
one unit is defined as the amount of enzyme required to incorporate 10nmoles of dNTPs into an acid precipitable form after 30 minutes at 70°C. |
COMMENTS: purified free of endonucleases ; performance was confirmed by using Lambda DNA as template (amplified fragment 7.5kb) and human placenta DNA as template (amplified fragment 3.0kb). Optimization efforts are often required. |
