M-MuLV REVERSE TRANSKRIPTASE - RNAse H minus MODIFIED

Moloney Murine Leukemia Virus (M-MuLV) Reverse Transkriptase is isolated from an E.Coli strain that carries a plasmid with the cloned modified M-MuLV RT gene with deleted RNAse H coding part. M-MuLV RTase is an RNA-dependent DNA polymerase. This enzyme can synthesize a complementary DNA strand initiating from a primer using either single-stranded RNA or DNA template. The enzyme lacks RNAse H activity.

CONCENTRATION:

    100 units/µl

REACTION BUFFER 10X:
  • 50 mM Tris-HCl pH 8,3
  • 2-4 mM MnCl2
  • 2-8 mM MgCl2
  • 10 mM DTT
  • 100mM KCl
  • 2 mM dNTP
  • Incubate at 37oC.
UNIT DEFINITION:

    one unit is defined as the amount of enzyme required to incorporate 1 nmol of dTTP into an acid precipitable form after 10 minutes at 37oC using poly(rA) -p(dT)10-20 as template/primer.

COMMENTS:

    tested for the ability to synthesize full length cDNA from 4 kb RNA.

    50 mM Tris-HCl pH 8,3, 1 mM EDTA, 0,1 mM DTT, 0,1mM NaCl, 0,1% Triton X-100, 50% glycerol.
