GENAXIS™ Extra Long PCR Kit is a unique system designed specially to produce a high yield of PCR products from various sources of DNA. It consists of a controlled mix of enzymes that can be used in any experiment where quantity, fidelity and long fragment size are required. It may be used to replace Taq DNA Polymerase and Pwo DNA Polymerase. However, the main application is for amplification of very long target sequences, including genomic DNA. The GENAXIS™ Extra Long PCR Kit amplifies fragments of up to 30 kb from human genomic DNA and up to 40 kb from lambda DNA.With this new methodology, the rapid characterization and cloning of complete genes from genomic DNA is possible, using cDNA sequence information as a start point.

APPLICATIONS:  

  • all standard PCR* reactions replacing Taq DNA Polymerasel
  • amplification of very long DNA sequences including genomic DNAl

KIT CONTENT:

    • 100 units enzyme mixture     (20µl)
    • 10X reaction buffer     (1ml)
    • 25mM MgCl2     (1ml)
    • Dilution buffer     (1ml)    

CONTENTRATION:

    5 unit/µl.

 REACTION BUFFER 10X :

 

    500mM Tris-HCl pH8.5, 140mM (NH4)2SO4 ; 25mM MgCl2 is supplied separately.
UNIT DEFINITION:

    one unit is defined as the amount of enzyme required to incorporate 10nmoles of dNTPs into an acid precipitable form after 30 minutes at 70°C.

 COMMENTS:

    purified free of endonucleases . The amplification is very robust up to 15kb (genomic DNA). Amplification of larger fragments is more critical for set up, template quality, denaturation conditions, correct conditions for dNTPs/Mg2+ concentrations.

 STORAGE AND DILUTION BUFFER:

    20mM Tris-HCl pH8.0, 100mM KCl, 0.2mM EDTA, 1mM DTT, 55% (v/v) glycerol and stabilizers. If necessary the enzyme can be diluted before use. The diluted enzyme may be stored at –20°C for up to six months without loss of activity.
