Transcriptase is isolated from the Avian Myeloblatosis Virus as the ab holoenzyme with a
molecular weight of 157,000 daltons. AMV Reverse Transcriptase has both a 5 ½
3 primer-dependent polymerase activity which uses either RNA or DNA as a template,
and a 3 ½ 5 RNase H activity that degrades the RNA from RNA/DNA hybrids. It
requires a primer as well as Mg2+ or Mn2+ ions. GENAXIS Biotechnology AMV Reverse
Transcriptase can be applied to dideoxy sequencing of DNA and RNA and to cDNA synthesis
using smaller RNA templates. In cDNA reactions, the enzyme is compatible with 4mM sodium
pyrophosphate and placental RNase inhibitor. The optimal temperature is 41-45°C. A
temperature of 37-41°C is preferable if NaPPi is used, however. GENAXIS
